体外研究 | 天蚕素A显示出抗癌活性。 Cecropin A(10-50μM)剂量依赖性地降低HL-60细胞的活力。 Cecropin A(30μM)促进ROS产生,导致线粒体膜电位(Δψm)崩溃,并在HL-60细胞中产生核染色质的形态变化。 Cecropin A(30μM)也可导致早期凋亡,并导致HL-60细胞中不依赖caspase的细胞死亡[1]。天蚕素A对革兰氏阴性细菌具有细胞毒性,包括鲍氏不动杆菌(CCARM 12005,CCARM 12035,CCARM 12036,CCARM 12037),其最小抑制浓度(MIC)为0.5-1μM。 Cecropin A(25μM)显着阻断mTNF-α,mIL-1β和mMIP-2 mRNA的表达,并略微抑制RAW264.7细胞中mMIP-1 mRNA的表达。 Cecropin A(0.1,0.25,0.5,1,2.5,5μM)也抑制NO的产生并降低LPS刺激的RAW264.7细胞中的mTNF-α细胞因子水平,并且具有抗炎活性[2]。 |
细胞实验 | 简而言之,将补充有10%热灭活FCS的RPMI 1640中的5×10 5个细胞/ mL置于96孔板上。将天蚕素A加入细胞培养物中,终浓度为10,20,30,40和50μM,将细胞在37℃,5%CO 2的潮湿气氛中培养24小时。然后,向每个孔中加入20μLMTT(0.5mg / mL),并将板在37℃下温育4小时。除去MTT溶液,向每个孔中加入含有0.04N盐酸的异丙醇以溶解甲crystals晶体。在分光光度微板读数器上在550nm的测试波长和620nm的参考波长下测定吸光度。在不存在天蚕素A(未处理的细胞)的情况下孵育的细胞的吸光度设定为100%。结果表示为细胞活力的百分比[1]。 |
